Procedure

  1. Peel a thin layer from the inner surface of an onion scale.
  2. Place the peel on a clean glass slide.
  3. Add a drop of safranin stain and wait 2 minutes.
  4. Place a cover slip gently, avoiding air bubbles.
  5. Observe under low power (10x) then high power (40x).
  6. Identify cell wall, cell membrane, nucleus, and vacuole.
