Apparatus
- Microscope
- Fresh leaf (Tradescantia / Rhoeo)
- Glass slide and cover slip
- Safranin stain
- Needle and forceps
- Dropper and blotting paper

Procedure
- Fold the leaf and gently peel the lower epidermis.
- Place the peel on a slide with a drop of water.
- Add safranin stain, wait 2 minutes, then place cover slip.
- Observe under 10x then 40x magnification.
- Identify guard cells, stomatal pore, and epidermal cells.
- Count stomata in the field of view.
