Apparatus

  • Microscope
  • Fresh leaf (Tradescantia / Rhoeo)
  • Glass slide and cover slip
  • Safranin stain
  • Needle and forceps
  • Dropper and blotting paper

Procedure

  1. Fold the leaf and gently peel the lower epidermis.
  2. Place the peel on a slide with a drop of water.
  3. Add safranin stain, wait 2 minutes, then place cover slip.
  4. Observe under 10x then 40x magnification.
  5. Identify guard cells, stomatal pore, and epidermal cells.
  6. Count stomata in the field of view.
